SOP for the operation of colony counter

I     PURPOSE

             To describe the procedure for the operation of colony counter

 

II   SCOPE

           This procedure is applicable to the Colony counter in Microbiology Department...

V.        PROCEDURE

1.1               Ensure that the instrument and working area are clean.

1.2              Ensure that the magnifying lens is fixed at the appropriate position.

1.3              Ensure that the marker pen is connected to the instrument.

1.4              Connect the instrument to the main power.

1.5              Switch “ON” the mains.

1.6              The internal light will be flashed on and digital screen displays “ 0000”.

1.7              Then observe the plate through the magnifying lens adjusted a distance from the object and count the no. of colonies by pressing the nib of the marker on each colony.

1.8              The display will show the no. of pressings i.e. no. of colony forming units.

1.9              Note down the counts.

1.10          Then press the RESET button to get the “0000” on the digital screen.

1.11          Switch off the mains.
