Method
for the estimation of Morindin content
Reagent
Required:
Methanol L.R. Grade, Sulphuric acid, Ammonia
solution.
Glass
wares and Apparatus Required:
250mL Conical flask, Gooch crucible, 50mL
Stoppered cylinder and Evaporating dish.
Procedure:
Weigh accurately about 5.0gm of dry extract powder
into a 250mL RB flask, Add about 25mL of 90% methanol . Reflux for 30minutes. Cool
& filtered through whatman No.1.Repeat the same two times more. Remove
solvent by distillation up to soft mass. Cool & reflux successively with 25mL each of Petroleum
ether, chloroform, carbon tetrachloride & ethyl acetate. Cool & discard the organic layer by
decantation. Dissolve the residual extract in 25mL of methyl alcohol. Filter through
whatman filter paper & concentrate up to 5mL.. Filtrate is slowly added with
constant stirring to 25mL of acetone into a preweighed dish. Left overnight to
get precipitate. Decant the supernatant liquid & discard. Dry the
precipitate at 105°C to constant weight
& weigh.

Percentage of Morindin
content is calculated as:
(A – B) X 100
