for the estimation of Andrographolides
content
Reagent
Required:
Methanol, Benzene, Ethyl acetate (LR Grade)
Glass
wares and Apparatus Required:
R.B. flask, condenser, soxhlet apparatus,
funnel, filter paper No. 1, glass rod, Evaporating dish.
Procedure:
Weigh accurately about 3.0gm of dry extract
powder in a small soxhlet apparatus with 200mL of methanol. Evaporate the
extract to 50mL on a water bath and add 30mL of water. Wash the aqueous
alcoholic extract with two 25mL portions of benzene and to the aqueous
alcoholic extract add 20mL of water and extract with four 25mL portions of
ethyl acetate. Combine the ethyl acetate extracts dry to constant weight
at 900 C and weigh the
andrographolide.
Calculation:
Percentage of andrographolide content (w/w)
is calculated as :
(A – B) X 100

For raw
material: 10gm
sample (20 #) is to be taken in the case of raw material and the quantity of
methanol shall be increased accordingly. Rest of the method is same as above.
