SOP FOR WASHING OF HPLC COLUMN

I.                   PURPOSE

To describe the procedure for washing of HPLC column.

This procedure is applicable to HPLC columns in Quality Control department,

V.                PROCEDURE

1.0              Instructions

1.1              After analysis, wash the column completely salt free.

1.2              Before storing, the column should be washed with 100% organic solvent    (Acetonitrile/ Methanol).

2.0       FOR C18, C8 and PHENYL COLUMNS

2.1       After usage , column should be washed with Milli-Q water at the flow rate of        1 ml/min for about 60 min.

2.2              After water washing, wash with 100% solvent (Acetonitrile / Methanol) at the flow rate of 1 ml/min for minimum 30 min.

2.3              Check that the pressure does not exceed 3000 PSI.

2.4              Enter the column usage details in ‘HPLC column usage log’

            Format No.: F01/SOPQC034-00.

2.5              Fix the end fittings properly and store in an appropriate place.

2.6              Before every use, wash the column with milli-Q water at the flow rate of 1ml/min for about 30 min.
